What this study did
Wagner et al. (2011) mailed surveys to 542 parents. Each parent had a child with one of six rare genetic syndromes. Parents filled out the Social Communication Questionnaire and two checklists about mood and activity.
The team wanted to map how autism traits, affect, and hyperactivity cluster in each syndrome.
What they found
Cornelia de Lange (CdLS) and Fragile X (FXS) groups scored highest on autism symptoms.
Kids with Smith-Magenis (SMS), Angelman (AS), CdLS, and FXS were rated as very impulsive.
Prader-Willi (PWS) children showed the lowest over-activity levels.
Each syndrome carried its own behavioral fingerprint.
How this fits with other research
DeFulio et al. (2011) looked only at cri-du-chat syndrome, one of the same six syndromes. They found that tired kids scored even higher on autism checklists. Chris et al. did not measure fatigue, so the two studies slot together: fatigue may boost the already high autism scores seen in CdCS.
D'Agostino et al. (2025) surveyed moms of FXS and ASD kids. They found that FXS mothers use more mindful parenting, yet still feel hassled. Chris et al. showed why: FXS children present strong autism and hyperactivity traits, giving moms plenty to manage.
Saghazadeh et al. (2017) pooled blood data and confirmed that idiopathic autism shows modest BDNF elevation. Chris et al. remind us that genetic-subgroup autism can look different, so biomarker studies need to split, not lump, these kids.
Why it matters
If you assess a child with CdLS or FXS, expect a heavy autism trait load and plan dense teaching trials. Watch for impulsivity in SMS, AS, and CdCS; use frequent breaks and response-cost. For PWS, keep sessions calm and seated. Always ask parents about fatigue—it can inflate scores and mislead eligibility decisions.

We investigated autism spectrum disorder (ASD) symptomatology, hyperactivity and affect in seven genetic syndromes; Angelman (AS; n = 104), Cri du Chat (CdCS; 58), Cornelia de Lange (CdLS; 101), Fragile X (FXS; 191), Prader-Willi (PWS; 189), Smith-Magenis (SMS; 42) and Lowe (LS; 56) syndromes (age range 4-51). ASD symptomatology was heightened in CdLS and FXS. High levels of impulsivity were seen in SMS, AS, CdCS, FXS and adults with CdLS. Negative affect was prominent in adults with CdLS, while positive affect was prominent in adults with AS and FXS. Heightened levels of overactivity and impulsivity were identified in FXS, AS and SMS while low levels were identified in PWS. These findings confirm and extend previously reported behavioral phenotypes.
